CCL8 (C-C motif chemokine ligand 8), also known as MCP-2, is a chemotactic cytokine, which was mainly produced in monocytes/macrophages, fibroblasts, endometrial cells and mast cells. CCL8 can exert a chemotactic effect on monocytes, T lymphocytes, NK cells, basophils and eosinophils.

Biocytogen developed a CCL8 humanized mice, the exons 1~3 of mouse Ccl12 gene that encode the full length protein were replaced by human CCL8 exons 1~3 in B-hCCL8 mice. Then, confirmed that human CCL8 protein was exclusively detectable in serum of homozygous B-hCCL8 mice.

Protein expression analysis
Strain specific CCL8 and CCL12 expression analysis in wild-type C57BL/6N mice and homozygous humanized B-hCCL8 mice by ELISA. Serum was collected from wild-type mice (+/+) and homozygous B-hCCL8 mice (H/H) stimulated with 40 μg LPS in vivo for 2 hours (male, 25-week old, n=3). Expression level of hCCL8 and mCCL12 were analyzed by ELISA (anti-mouse CCL12 ELISA kit: Abcam, ab246547; anti-human CCL8 ELISA kit: Abcam, ab223856). Mouse CCL12 was only detectable in wild-type C57BL/6N mice. Human CCL8 was exclusively detectable in homozygous B-hCCL8 mice but not in wild-type mice. Values are expressed as mean ± SEM.
